The conventional methods of amino acid analysis had the following difficulties.

  • Post-labeling Methods: Utilizes ion-exchange modes, requiring complete separation of amino acids, leading to complex mobile phases and difficulties in shortening analysis time.
  • Pre-labeling Methods: Allows for short-time analysis using reversed-phase modes, but requires time and equipment for amino acid pre-labeling reactions. Additionally, labeling efficiency is susceptible to sample matrix interference.
